Molecular dissection of integrin signalling proteins in the control of mammary epithelial development and differentiation.
Cell-matrix adhesion is essential for the development and tissue-specific functions of epithelia. For example, in the mammary gland, beta1-integrin is necessary for the normal development of alveoli and for the activation of endocrine signalling pathways that determine cellular differentiation. However, the adhesion complex proteins linking integrins with downstream effectors of hormonal signal...
متن کامل2-methoxyestradiol induces mammary gland differentiation through amphiregulin-epithelial growth factor receptor-mediated signaling: molecular distinctions from the mammary gland of pregnant mice.
Levels of 2-methoxyestradiol (2ME(2)), an endogenous metabolite of estradiol, are highly elevated during late stages of pregnancy when mammary glands have differentiated with the formation of alveolar structures producing milk proteins. Based upon our previous demonstration that 2ME(2) induces mammary ductal dilation associated with expression of mammary differentiation markers when administere...
متن کاملMammary gland development and cancer.
The mammary gland is a complex organ that begins development early in gestation and constantly changes in size, shape and function from the time of puberty to menopause. The earliest stages of embryogenesis appear to be independent of steroid hormones, whereas after the 15th week breast structure is largely influenced by a variety of hormones. In most females, further breast development begins ...
متن کاملMolecular and cellular basis of the mammary gland susceptibility to carcinogenesis
Mammary carcinomas induced by the administration of 7,12-dimethylbenz(a)anthracene (DMBA) to young virgin rats arise from undifferentiated terminal ductal structures called terminal end buds (TEBs). TEBs that normally differentiate into alveolar buds (ABs) and lobules under the influence of DMBA develop intraductal proliferations which progress to carcinoma. The high susceptibility of the young...
